Phytofabrication of biomolecule – coated metallic AgNps using leaf extracts of Nyctanthes Arbor-tristis and its antibacterial & anticancer activity against human MDA-231 cell line

Abstract

The present study reports an eco-friendly path and cost-effectiveness featured Silver Nanoparticles (AgNPs) from leaf extract of Nyctanthes arbor-tristis L. Phytochemical characterization administrated qualitatively for the presence of alkaloids, flavonoids, carbohydrates, saponins, glycosides, steroids, tannins, terpenoids and protein. AgNPs synthesis was confirmed by visual confirmation as color change and the synthesized AgNPs were examined using various characterization techniques includes UV–Visible spectroscopy, X-ray diffraction (XRD), Energy Dispersive X-ray spectroscopy (EDX), Fourier Transform Infrared Spectroscopy (FTIR), Scanning Electron Microscope (SEM) and Dynamic Light Scattering (DLS). Furthermore, antibacterial potential of AgNPs was assessed against bacterial strains such as Escherichia coli, Pseudomonas aeruginosa, Salmonella typhi, Staphylococcus aureus and Klebsiella pneumonia and the maximum Zone of inhibition was observed against Salmonella typhi (24.6mm), Staphylococcus aureus (21.6±0.57mm), Pseudomonas aeruginosa (19mm), Klebsiella pneumonia (17mm) and Escherichia coli (15.3±0.57mm). Synthesized AgNPs also possess anticancer properties with MDA-231 cell line (Human Breast cancer cell line) which were determined with dose-dependent manner with the concentration of 2.5 to 30 μg/mL respectively. The inhibition range was observed as the concentration increases at 30 μg/mL. In this work, it has been investigated that high concentration of leaf extract possess high inhibitory potentials in both bactericidal and cytotoxic assays.
